Gene walking involves the complete sequencing of large (>1kb) stretches of DNA. Beginning from your reference sequence, stage 1 of sequencing is started, then onwards, primers are designed to perform overlapping sequencing of the remaining DNA and assemble the sequence data.  Additionally, if you have a known sequence for which you need quality resequencing information, a complete set of overlapping primers will be designed for generating complete sequence
